My first test yesterday w/ USB Loader using card reader (specifically USB 2.0 SD MMC Reader USB Device <PnPName=USB\VID_14CD&PID_8123\812822222789> bought from CDRKing) and 4GB PQI SDHC (also from CDRKing)
The GUI I used was from calimero100582 a 'real' wbfs windows port - GBAtemp.net. There are now lots of GUIs for Windows, Linux and Mac(? - I think). I just found that particular GUI easy to do batch uploading. Also, if you're more comfortable using CLI, then good for you. It's really the user's preference.
From the test, you can see it was a successful added to the WBFS partition of my test usb device.
When I test played Don King Boxing (NTSC/U) release on my 3.2u Wii, I the loading times faster than playing from disc.
For more developments related to Wii USB ISO Loader, I recommend you lurk around gbatemp forums ;D
Add: The ISO I used was untouched (not scrubbed) BTW.

What I've test installed so far on the Buffalo MiniStation Lite:
It's now possible to load games inside wbfs directly from the main wii menu via channels ! ;D

Update: WBFS Manager and Channel Creator updated to 1.5 and it works great! I can easily make channel wads for any games I've loaded to my hdd ;D
